A simple method is described for the separation of ectoderm from endoderm in sea urchin pluteus larvae. The separation, by sequential treatments with glycine-EDTA, permits an examination of the basement membrane beneath the ectoderm layer and of a fibrous layer beneath the basement membrane. In a serological study the two germ layers are shown to differ with respect to cell surface antigen composition. Further, the basement membrane is shown to contain ectoderm antigens.
